Gut Bacteria Turn to Mucus Lining When Fibre Is Scarce

Gut bacteria may start breaking down the protective mucus lining of the intestine when dietary fibre runs short, according to two studies published in the Proceedings of the National Academy of Sciences and Nature Metabolism by researchers at the Ludwig Institute for Cancer Research at Princeton. Using stable isotope tracking in mice, a team led by Jenna AbuSalim and director Joshua Rabinowitz found that when fibre was scarce, gut microbes turned to consuming proteins in the gut's mucus barrier, producing higher levels of phenol compounds that other research has linked to poorer outcomes in people with kidney disease and some cancers. Fibre appeared to suppress that process. The researchers also identified a class of indigestible plant proteins they call Prif, short for "proteins imitating fiber", which shifted bacterial metabolism toward more beneficial compounds instead. The work, funded by the Ludwig Institute and the US National Institutes of Health, was first announced by EurekAlert on 27 July 2026 and covered again this week by ScienceDaily.

Why this complicates gut health research

The findings challenge the assumption that gut bacteria's byproducts come only from bacteria themselves. The researchers showed mammalian cells can independently produce many of the same metabolites, complicating how such compounds are interpreted in gut health studies. Rabinowitz said the team wants to understand "what aspects of the diet control which microbial outputs" before the findings can inform dietary guidance. The current evidence comes from mouse models, cell cultures and samples from cancer patients on antibiotics; a controlled human dietary trial has not yet been published, and AbuSalim described Prif as "an emerging class of dietary nutrients" whose broader health effects are still being mapped.
